My friend has a knot on one of his testicles, is it a possibility that he has a sexual transmitted disease?I would like the answer from a man or a medical physician.

Unlikely that it is an STD. However, while there are benign (harmless) conditions that can cause knots in the testicles, there are also some dangerous (for him...not you) conditions. For that reason, I'd encourage you to encourage him to see a doctor.

If it is on the skin, it cld be a cyst eg - sebaceous cyst. Inside the scrotal sac, it could be a varicocele, inflammation of epididymis or any growth. Best will be to get a check up. It should not be an STD.
